Proliferation of astrocytes in hippocampus of mice following middle cerebral artery occlusion
Abstract:Objective To study the proliferation of astrocytes in hippocampus of mice following middle cerebral artery occlusion (MCAO).Methods Sixty-four Kunming mice were randomly divided into cerebral ischemia group (n=48),normal group (n =4),and sham operation group (n =12).A MCAO model of micc was established by Langa occlusion.The ischemic site and size were observed with TTC staining,the proliferation of astrocytes in hippocampus was assayed with HE staining,the expression of GFAP was detected with immunohistochemical staining at 1,3,6,12,24 and 48 h after ischemia.Results The infarction size was 19.67%± 5.46 %,33.68 % ± 3.46%,54.65 % ± 6.13 %,76.85 % ± 7.15 %,81.81 % ± 4.09 % respectively in cerebral ischemia group at 3,6,12,24 and 48 h after ischemia,which was significantly larger than that in normal group (P=0.000).HE staining showed that the damage of neurons in hippocampus exacerbated and became irrepairable in cerebral ischemia group at 6 h after ischemia.The expression level of GFAP was significantly higher in cerebral ischemia group than in normal group and sham operation group (P<0.05).The cell body was swollen,the protrusion became thicker and longer,the number of GFAP-positive cells reached its peak at 6 h after ischemia (P<0.05).Conclusion MCAO can damage the neurons in hippocampus and stimulate the proliferation of astrocytes in mice.
