Effect of recombinant human relaxin on diastolic function in a rabbit diastolic heart failure model
Abstract:Objective To study the effect of recombinant human relaxin (rh-RLX) on diastolic function in a rabbit diastolic heart failure (DHF) model.Methods Six out of the 30 New Zealand rabbits served as a sham operation group.A DHF model of 30 rabbits was established by abdominal aorta constriction.The animals underwent echocardiography at week 8 after operation and were divided into DHF group (n=6),benazepril (3 mg/kg · d) group (n=8),low rh-RLX dose (30 μg/(kg · d) group (n=8),high rh-RLX dose (98 μg/kg · d) group (n=8).After 1 week of drug treatment,their diastolic function was assayed by echocardiography and their myocardial fibrosis was detected with Masson staining at week 9 after operation.Results The E/A was significantly higher,the LVPW and IVS were significantly thinner,the LVEDP was significantly lower in high rh-RLX dose group than in DHF group (1.36±0.10 vs 0.83±0.06,P<0.01;3.71±0.21 mm vs 4.87±0.08 mm,P<0.01;3.10±0.08 mm vs 3.23±0.08 mm,P<0.05;8.74±1.16 mm Hg vs 12.07±1.31 mm Hg,P<0.01).The ratio of E/A was significantly higher,the LVPW was significantly thinner,the LVEDP was significantly lower in high rh-RLX dose group than in benazepril group (P<0.05,P<0.01).Conclusion High rh-RLX dose improves diastolic function in DHF rabbits by inhibiting their myocardial fibrosis.
