Effect of resveratrol on autophagy of aged myocardium through the Sirtuin 1-Beclin 1 signaling pathway
Abstract:Objective To study the effect of resveratrol on autophagy of aged myocardium and its molecular mechanism.Methods Eighteen C57BL/6 mice were divided into 4 months old group,22 months old group,resveratrol-treated 22 months old group (6 in each group).LC3 protein expression was detected by Western blot,Beclin1 acetylation was assayed by immunoprecipitation,and SIRT 1 activity was measured by fluorescent quantitation in 3 groups.The number of GFP-LC3 particles in cultured H9C2 myocytes transfected with GFP-LC3 adenovirus was calculated after the Beclin 1 gene was interfered with siRNA.Results The relative activity of SIRT1 was 100.00%±6.21%,74.60 %±7.85%,112.30%±8.43% respectively and the expression level of LC3 Ⅱ/LC3 Ⅰ was 1.000±0.046,0.524±0.038,0.837±0.044 respectively in 3 groups (P<0.05).The relative activity of SIRT1 and the expression level of LC3 Ⅱ/LC3 Ⅰ were significantly lower in 22 months old group than in 4 months old group and were significantly higher in 4 months old group than in resveratrol-treated 22 months old group (P<0.05).The Beclin1 acetylation was higher in 22 months old group than in 4 months old group,which could be downregulated by resveratrol.The number of GFP-LC3 particles in H9C2 myocytes increased after the Beclin1 gene was interfered with siRNA with no change occurred after resveratrol was added.Conclusion Resveratrol can increase the autophagy of aged myocardium through the SIRT1-Beclin1 signaling pathway.
