Effect of cerebral ischemia preconditioning on antioxidant stress pathways in rats following I/R injury
Abstract:Objective To study the effect of cerebral ischemia preconditioning (CIP) on Nrf2 and γ-GCS pathways in rats following I/R injury.Methods One hundred and eighty male SD rats were randomly divided into sham operation group,model group,and CIP group (60 in each group).The anaimals were killed after their neurological function was scored at 6,12,24,48 and 72 h.Expressions of Nrf2 and γ-GCS mRNA in cerebral cortex were detected by RT-qPCR with immunohisto chemical staining and serum GSH level in cerebral tissues was measured.Results The behavioral score was significantly higher in model group and CIP group than in sham operation group at different time points (P<0.01) and was significantly lower in CIP group than in model group at 24,48 and 72 h (2.00±0.63 vs 2.83±0.41,2.16±0.41 vs 2.83±0.41,1.17±0.41 vs 2.00±0.89,P<0.05,P<0.01).The number of positive cells during Nrf2 nuclear shift was significantly grea ter,the expression levels of Nrf2 and γGCS mRNA,and the serum GSH level were significantly lower in model group and CIP group than in sham operation group (P<0.05,P<0.01),and were significantly higher in CIP group than in model group (P<0.05,P<0.01).Conclusion CIP regulates the expression of downstream oxidation stress products and protects aganst I/R injury-induced cerebral tissue damage through the Nrf2/γ-GCS signaling pathways.
