Effect of 3-n-butylphthalide on blood brain barrier in rats following I/R injury
Abstract:Objective To study the effect of 3‐n‐butylphthalide (NBP ) on blood brain barrier (BBB) in rats following I/R injury .Methods Seventy‐two male SD rats were randomly divided into sham operation group ,model group ,and NBP treatment group (24 in each group) .A rat I/R injury model was established by Longa occlusion .Water and Evans blue levels in brain tissue were measured 2 h after ischemia and 24 h after reperfusion .The morphology of BBB was observed by transmission electron microscopy .Expressions of MMP‐9 mRNA and TIMP‐1 mRNA in brain tis‐sue were detected by RT‐PCR .Results The water and Evans blue levels and the MMP‐9 mRNA and TIMP‐1 mRNA expression levels were significantly higher in model group than in sham oper‐ation group (79 .14% ± 1 .65% vs 73 .06% ± 0 .68% ,9 .45 ± 0 .94 μg/g vs 3 .45 ± 0 .45 μg/g ,P<0.01) .The water and Evans blue levels and BBB injury extension level were significantly lower , the MMP‐9 mRNA expression level was significantly lower and the TIMP‐1 mRNA expression level was significantly higher in NBP treatment group than in model group (P<0 .01) .Conclusion NBP can alleviate BBB in rats following I/R injury by regulating the MMP‐9 mRNA and TIMP‐1 mRNA expression .
