Effect of drug intervention on connexin 43-mediated oxidative stress in sympathetic atrial fibrillation dogs
Abstract:Objective To study the effect of membrane protein transduction blockers and reactive oxygen species (ROS)scavengers on connexin 43 (Cx43)and oxidative stress ,and its relation with atrial fibrillation(AF) inducibility .Methods Twenty‐four China rural dogs were randomly divid‐ed into control group ,sympathetic AF group A ,sympathetic AF group B ,sympathetic AF group C (6 in each group) .Their atrial effective refractory period (AERP)and AF inducibility were tested before and after sympathetic stimulation and drug intervention .Total Cx43 protein and its phos‐phorylation level were measured .Results No significant change was found in total Cx43 protein and AERP between control group and 3 sympathetic AF groups (P>0 .05) .The Cx43 phospho‐rylation level was significantly lower in 3 sympathetic AF groups than in control group (P<0 .05) and in sympathetic AF group B than in sympathetic AF groups A and C (P<0 .05) .Conclusion Membrane protein transduction blockers can suppress the transport of cell membrane Cx43 to mi‐tochondrial membrane .Deceased Cx43 phosphorylation level can lead to oxidative stress imbal‐ance ,thus increasing the susceptibility of AF .ROS scavengers can reduce excessive oxidative stress‐induced Cx43 damage in cell membranes and mitochondria ,thus relieving atrial electrical remodeling and AF inducibility .
