Effect of flunarizine on Na+ current in rat cortical neurons after primary culture
Abstract:Objective To study the effect of flunarizine on voltage-gated Na+ channel current in rat cortical neurons and its mechanism .Methods Cortical neurons from 17 -18 d pregnant SD rats were cultured .Flunarizine (0 .1 ,1 .0 ,10 .0 ,100 .0 μmol/L) was added into the Na+ channel cur-rent using the whole cell patch clamp technique followed by 1 μmol/L .The Na+ current-associat-ed curves were compared before and after flunarizine was added .Results Flunarizine (at the con-centration of 0 .1 ,1 .0 ,10 .0 ,100 .0 μmol/L) inhibited the Na+ current in a concentration-depend-ent manner with an IC50 value of 0 .94 μmol/L .The shift of Na+ current inactivation curve to hy-perpolarizing direction was 17 .03 mV higher after flunarizine was added than before it was added . The Na+ channel recovery time increased to 19 .46 ± 0 .03 ms from 7 .51 ± 0 .05 ms (P<0.05) . The Na+ current decreased to 9 .44% ± 5 .13% and 12 .56% ± 6 .72% after flunarizine was added from 21 .63% ± 7 .36% and 45 .83% ± 10 .04% before flunarizine was added (P<0 .05) .Conclu-sion The inhitory effect of flunarizine on cortical neurons can explain why it can prevent mi-graine attack .
