Study on the in vitro induction of human umbilical cord blood-derived mesenchymal stem cells into neurons by different inducers
Abstract:Objective To study the effect of different inducers on the differentiation of human um-bilical cord blood-derived mesenchymal stem cells(MSCs) into neurocytes in vitro. Methods The MSCs of third passage were induced by three methods:the group of chemical induction;the group of growth factor induction;the group of tanshinol and growth factor induction. Neuronal charac-teristic markers(NeuN,β-TubulinⅢ) and glial fibrillary acid protein(GFAP) were detected by the immuocytochemical staining and immunofluorescence method before and after induction. Results MSCs displayed neuronal morphology and long processes were protruded from the cell body when the MSCs were induced by these three methods. The induced cells derived from MSCs expressed NeuN and β-TubulinⅢ, but few expressed GFAP. The rates of NeuN and β-TubulinⅢ positive cells in tanshinol and growth factor induction group were higher than those in the chemical induc-er group and the growth factor inducer group,while the rate of GFAP positive cells was lower than that in growth factor induction group,with significant differences. Conclusion The combina-tion of tanshinol,epidermal growth factor and basic fibroblast growth factor can effectively induce mesenchymal stem cells derived from human umbilical cord blood into neurons.
