Effect of sarcoplasmic reticulum calcium ATPase 2a overexpression on cardiomyocyte apoptosis induced by hypoxia
Abstract:Objective To determine the effect of sarcoplasmic reticulum calcium ATPase 2a (SERCA2a) overexpression on the cardiomyocyte apoptosis induced by hypoxia. Methods Neo-natal rat ventricular cardiomyocytes were randomly divided into four groups, control group, hy-poxia group,SERCA2a group,SERCA2a+hypoxia group. After transfection with SERCA2a gene for 48 h, cardiomyocytes were cultured in an airtight chamber gassed with 95% N2/5% CO2 at 37℃ for 12 hours. The flow cytometry was employed to assess cell apoptosis rate. SERCA2a and caspase-3 expression was detected by Western blot. Intracellular free Ca2+ concentration was measured by calcium fluorescent probe Fluo3/AM. Results Transfection resulted in up regula-tion of expression of SERCA2a(2.5-fold increase in comparison with control group). Compared with control group, the apoptosis rate,caspase-3 expression and free Ca2+ concertration in hypoxia group were increased significantly. Compared with hypoxia group, the above measurements were decreased significantly after SERCA2a overexpression. Conclusion SERCA2a overexpression can reduce hypoxic cardiomyocyte apoptosis by attenuating the overload of intracellular calcium and the activation of caspase-3.
