Abstract:Objective To preliminarily explore the toxicity of the new Ti-6Al-4V-3Cu antibacterial implant material implanted in New Zealand white rabbits, and to provide experimental evidence for the clinical experiment of this implant material. Methods Eighteen adult female New Zealand white rabbits weighted 2.5-3.0 kg were randomly divided into three groups including experimental group, control group and blank group containing six rabbits in each group, where the Ti-6Al-4V-3Cu(TC4-3Cu)antibacterial implant and Ti-6Al-4V(TC4)implant were respectively implanted into the experimental group and control group in the present work.Gross observations were conducted on days 1,3,7 postoperatively, and complete blood count and blood biochemistry were analyzed preoperatively and on days 1,3,7 of post-surgery.One month after surgery, the rabbits were euthanized, and tissue samples were collected from major organs, including the heart, lungs, liver, kidneys, and spleen.Hematoxylin-eosin staining was employed to evaluate cellular morphology in these organs across experimental groups. Results All the experimental animals exhibited the good wound healing on days 1,3,7 of post-surgery.The TC4-3Cu and TC4 implants showed the negative effect on and the relative expression of compared to the blank group. One month after implantation, histological analysis revealed that there was the normal cellular morphology in the target tissues including heart, lung, liver, kidney and spleen without the inflammatory cell infiltration. Conclusion A novel Ti-6Al-4V-3Cu antibacterial biomaterial implanted into the New Zealand white rabbits without acute toxicity in vivo.
