Abstract:Objective This study aims to analyze the differences between the microRNA(miRNA)of the jawbone and the long bone of rats under the action of zoledronic acid. Methods Male rats were selected and randomly divided equally into:the normal saline(NS)group and the zoledronic acid(ZA)group. After treating for 12 weeks, all rats were sacrificed after 4 weeks of discontinuation, and 4 groups of samples were obtained from the mandible and femur, and the expression differences were analyzed by histological staining,miRNA sequencing, functional enrichment, and qRT-PCR on the bones of the four groups. Results The jawbone in the ZA group showed early osteonecrosis than the NS group.Under the action of ZA,miRNAs underwent specific changes in the jawbone were mainly related to the synthesis, secretion and function of parathyroid hormone, nuclear factor kappa-B signaling pathway, and apoptosis. Conclusion Under the action of zoledronic acid, jawbone histological osteonecrosis manifests earlier than clinically, and some miRNAs only undergo specific changes in the jawbone and may be related to BRONJ occurrence.
