Abstract:Cuproptosis as a novel form of regulated cell death,is characterized by copper ion accumulation, which triggers oxidative stress and metabolic disturbances. This process exhibits considerable potential for tumor prevention and treatment. This review systematically elucidates the roles of copper metabolism and cuproptosis in physiological processes, the relationship between cuproptosis and tumors, and particularly focuses on the mechanism of action, current research status, and clinical translational prospects of cuproptosis in oral squamous cell carcinoma(OSCC).Dysregulated copper metabolism can facilitate tumor development, whereas the cuproptosis mechanism exerts its anti-tumor effects by targeting high-metabolic tumor cells to inhibit proliferation, migration, and drug resistance.OSCC studies reveal that modulation of cuproptosis-related molecules can influence cellular behavior and is associated with risk factors.Compared to other squamous cell carcinomas,OSCC is more susceptible to environmental factors influencing copper homeostasis.By integrating in vitro and in vivo evidence, this review aims to provide novel perspectives for the diagnosis, treatment, and prognostic evaluation of OSCC.
