Effect of intermittent injection of parathyroid hormone on the vascularization of osteoporotic bone tissue

Abstract:Objective To investigation.The histological distribution of blood vessels and perivascular cells in the bone of mice following intermittent administration of parathyroid hormone(PTH).Methods Twelve 8-week-old male C57BL/6J mice were randomly allocated into two groups:a control group and a PTH-treated group,with six mice in each group.The control group received intraperitoneal injections of 0.9%saline,whereas the PTH-treated group received intraperitoneal injections of human parathyroid hormone[hPTH(1-34)]daily for 14 days.Following the treatment period,the femurs and tibias of the mice were harvested for tissue protein microarray analysis and immunohistochemical staining.Results The heat map results showed significant changes in proteins involved in multiple biological processes such as angiogenesis and intercellular communication in the long bones of PTH group mice.Compared with the control group,the number of endothelial mucin(+)/EphB4(+)venous system vessels and α-smooth muscle actin(+)perivascular cells in PTH-treated mice increased.The expressions of endothelial mucin,α-smooth muscle actin,EphB4,ephrinB2 in bone tissue vascular endothelium and perivascular proteins,as well as hypoxia-inducible factor 1α,c-kit,and kinase insert domain receptor in perivascular cells were all significantly upregulated(P<0.05).Conclusion Intermittent PTH(1-34)administration may enhance bone mass by influencing bone-specific blood vessels and perivascular cells.
