Culture and identification of dental pulp stem cells from human inflammatory tissue of deciduous teeth

Abstract:Objective To investigate the feasibility of isolating stem cells from inflamed pulps of deciduous teeth (SCID) derived from human inflammatory tissue,and to identify the function of the isolated cells.Methods The inflammatory pulp,obtained through the pulpotomyprocedure from 6 patients with pulpitis in deciduousteeth,was processed for SCID isolation. The primary SCID was cultured until P5,and the SCID was identified as mesenchymal stem cells (MSCs). Results SCID were successfully extracted from inflamed deciduous dental pulp without bacterial contamination. SCID showed good cell morphology,clonal formation and proliferation ability of MSC,and did not show obvious senescence phenotype after cell culture to P5. SCID could differentiate into osteogenic,adipogenic,and neurogenic lineages under induction culture conditions. Flow cytometry confirmed the expression of MSC markers CD73,CD90,and CD105,as well as the absence of hematopoietic markers CD34 and CD45 in SCID. Meanwhile,SCID cells injected into mouse tail veins caused no significant changes in blood routine tests. Conclusion Pulpotomy can preserve a part of SCID with MSC characteristics in infected pulp,which initially verifies the good biocompatibility of SCID and provides a possibility for further research on the functional characteristics of stem cells in inflammatory conditions and clinical translation application. It also broadens the source of dental pulp stem cells in milk teeth.
