Role of Notch pathway in RANKL/OPG expression induced by Porphyromonas gingivalis Lipopolysaccharidein human periodontal ligament cells

Abstract:Objective:To detect the expression of Notch signaling factors in human periodontal ligament cells(hPDLCs) treated with Porphyromonas gingivalis lipopolysaccharide (Pg-LPS),and further explore their roles involving in the expression of RANKL/OPG.Methods:Primary hPDLCs were cultured using combined methods of tissue explant and enzymatic digestion.Passage third-fifth cells were treated with 0.1、1、10μg/ml Pg-LPS,and the expression of RANKL/OPG in hPDLCs was detected by Real-time PCR after 72h.Meanwhile,the expressions of DLL-1,Hey-1,Notch4,Jagged1-2,Notch1-2 were assessed in hPDLCs treated with 1 μg/ml LPS.Furthermore,hPDLCs were pretreated with the γ-secretase inhibitor,namely DAPT,for 1 hour,and then co-cultured with 1 μg/ml Pg-LPS for 72 hours,the expression of RANKL/OPG and Hey-1 were measured using western-blotting and real-time PCR.Results:The mRNA expression of RANKL,as well as DLL-1、Jagged-1、Notch4 and Hey-1 were significantly elevated in hPDLCs stimulated with 1 μg/ml Pg-LPS (P<0.05).After blocking the notch signaling pathway with DAPT,both mRNA and protein levels of RANKL and Hey-lexpression were down-regulated as comparing with control hPDLCs treated with Pg-LPS (P<0.05).Conclusion:Notch signaling pathway is involved in the Pg-LPS induced RANKL/OPG expression in human periodontal ligament cells.
