Experimental study of subcutaneous osteogenesis by dog bone marrow stromal cells with nHAC/CSH

Abstract:Objective:T he aim of the study w as to assess osteogenesis of nano-hydroxyapatite/collagen/calcium sulphate hem ihydrate (nH A C/C SH ) loaded w ith dog bone m arrow strom al cells (dB MSC s) in nude m ice in vivo. Methods: D og B MSC s w ere isolated from healthy adultdogs by density gradientcentrifugation.nH A C/C SH w as m ixed w ith the induced cells and the constructs w ere exam ined under scanning electron m icroscopy (SE M). T he constructs w ere subcutaneously injected into the back ofnude m ice as experim entalgroup,and the nH A C/C SH as controlgroup. H&E staining w as used to exam ine the bone form ation at 4 w eeks. Results: SE Mshow ed the attached dB MSC s had a spread m orphology w ith polygonalshapeonnHAC/CSH.HE stainingshowedbone-liketissuefor mation 4 weeks later in the experimental group. Conclusion:T he com plex constructed w ith dB MSC s and nH A C/C SH show s good osteogenetic activity.
