Effect of tea polyphenols on apoptosis and inflammatory cytokines of human periodontal ligament fibroblasts induced by lipopolysaccharide

Abstract:Objective:To evaluate the effect of tea polyphenols on apoptosis and expression of tumor necrosis factor-α(TNF-α)and interleukin-6 (IL-6)in LPS-induced human periodontal ligament fibroblasts(HPDLFs).Methods:HPDLFs were cultured in vitro, and treated with LPS(1μg/ml)or LPS(1μg/ml) plus different concentrations of tea polyphenols(10μg/ml, 50μg/ml, 100μg/ml), respectively. Apoptosis was determined by flow cytometer, and the expressions of TNF-αand IL-6 were determined by Elisa.Results:Apoptosis was markedly enhanced and the expression of TNF-αand IL-6 were significantly up-regulated in HPDLFs stimulated by LPS. Tea polyphenols significantly inhibited LPS-stimulated apoptosis, as well as reduced the expression of TNF-αand IL-6.Conclusion:Tea polyphenols can effectively reduce apoptosis and the expression of TNF-αand IL-6 in LPS-induced HPDLFs.
