Exosome extraction and labeling for potential inner ear therapy

Abstract:Methods Umbilical cord mesenchymal stem cells were primary cultured and exosomes were isolated via ultracentrifugation.The isolated exosomes were labeled with fluorescence.Transmission electron microscopy,nanoparticle tracking analysis,western blotting,RNA extraction,and surface protein positivity assays were performed to characterize the exosomes.Results The isolated exosomes displayed a clear boundary,spherical morphology,and uniform size distribution.Nanoparticle tracking analysis showed an exosome particle concentration of 2.22×1010 particles/mL with a particle size of 81.97 nm.The bicinchoninic acid assay determined the exosomal protein concentration as 0.36 μg/μL.Characteristic exosomal membrane proteins,including TSG101,CD9,CD81,CD63 and Syntenin were detected.The RNA concentration of the exosomes was measured at 1.5 ng/μL.Conclusions This experimental method successfully isolated and purified exosomes for inner therapy.
