The Role of Glucose Transporter 3 in Noise-induced Hearing Loss

Abstract:Objective To report changes of glucose transporter 3(Glut3)at different time points after noise exposure.Methods C57BL/6J mice were selected and randomly divided into control group(10 mice)and noise exposure(NE)group(18 mice).In the control group,expression and localization of Glut3 in the cochlea were determined by Western blot,cochlea frozen sections,basilar membrane preparation and immunofluorescence staining.Mice in the NE group were subjected to white noise at 115 dB SPL for 4 h to induce permanent threshold shift(PTS),and sampled immediately,at 1 h or 24 h after noise(n=4 in each group).Cochlear basilar membrane sections with immunofluorescence staining were used to detect changes in Glut3 expression.The remaining 6 mice in this group were subjected to auditory brainstem response(ABR)tests at 7,14,21 and 28 d after noise modeling to verify successful modeling.Results Glut3 was widely expressed in cochlear tissues.Compared with the control group,the level of Glut3 expression was higher immediately after noise exposure.Conclusion Glut3 expression is significantly elevated immediately after noise exposure,likely playing a role of rapid response to noise and enhanced energy supply.
