Cisplatin-Induced Mitochondrial Function Damage in HEI-OC1 Cells

Abstract:Objective To demonstrate apoptosis of HEI-OC1 cells induced by cisplatin ototoxicity,and its effect on mitochondrial function.Methods HEI-OC1 cells were exposed to cisplatin,and the optimal concentration of cisplatin to cause cellular damage determined by CCK8.Apoptosis,mitochondrial membrane potentials,mitochondrial biosynthesis and mitochondrial oxidative stress were examined by flow cytometry and live cell staining.Reverse-transcriptase quantitative PCR(RT-qPCR)was used to determine transcription levels of mitochondrial biosynthesis and mitochondrially-encoded genes,including NRF-1,mtTFA,Tomm20,VDAC1 and COXV.Results Compared with the control,cisplatin at 10 μM/L induced 50%mortality in HEI-OC1 cells.HEI-OC1 cells showed lower mitochondrial membrane potentials,higher mitochondrial biogenesis and enhanced oxidative stress after cisplatin exposure,in parallel with enhanced transcription of mitochondrial biosynthesis and mitochondrial proteins genes(P<0.05).Conclusions In this in vitro study,treatment of HEI-OC1 cells with cisplatin caused damage to mitochondrial related functions,enhanced mitochondrial oxidative stress and resulted in apoptosis of HEI-OC1 cells.
