Ginkgolide B Reduces Tobramycin Induced HEI-OC1 Cells Apoptosis Via Regulating E2f1/Cdk1

Abstract:Objective To study the roles of Ginkgolide B in protection of House Ear Institute-Organ of Corti 1(HEI-OC1)cells from apoptosis induced by tobramycin and underlying mechanisms.Methods HEI-OC1 cells were treated with tobramycin with or without the presence of Ginkgolide B.Cell proliferation was measured by EdU assay and cell apoptosis by flow cytometry.Western blot assay was used to measure protein expression levels of apoptosis-re-lated genes.Results Proliferation of HEI-OC1 cells was suppressed by tobramycin(.P<0.001),with increased apoptosis(P<0.001).Western blot revealed decreased protein levels of Bcl2,and increased protein levels of Bax,Cleave-Caspase-3 and Cleave-Caspase-9(P<0.001).Adding Ginkgolide B to tobramycin treatment significantly decreased expression levels of E2f1 and Cdk1 and inhibited tobramycin-induced apoptosis(P<0.050).Conclusion Ginkgolide B may protect HEI-OC1 cells from apoptosis induced by tobramycin via regulating E2f1/Cdk1 pathway.
