Auditory Brainstem Responses in Nlrp3 Gene Knockout Mice

Abstract:Objective To evaluate hearing in Nlrp3 gene knockout mice at different months of age.Methods Thir-ty-two Nlrp3 knockout homozygous mice and 32 wild-type C57 mice were divided into 4 groups based on age,i.e.1,3,6 and 12 months.Auditory brain stem responses(ABRs)were detected using clicks and tone bursts(16 kHz),and outer hair cells were counted.Results In homozygous Nlrp3 knockout mice,ABR thresholds were 41.670±12.800 dB SPL,44.380±17.309 dB SPL,45.000±16.298 dB SPL and 60.000±6.200 dB SPL at 1,3,6 and 12 months,respectively,com-pared to 19.500±8.790 dB SPL,23.750±11.790 dB SPL,25.940±2.016 dB SPL and 41.670±4.710 dB SPL in wild-type mice,with threshold differences at 1 and 12 months being statistically significant(P<0.05).ABR results with tone burst(16k)were similar.Partial loss of outer hair cells was noticed in Nlrp3-/-mice,with less outer hair cells than in C57 mice.Conclusion Nlrp3 gene knockout leads to decreased number of outer hair cells and moderate to severe hearing loss in mice.Hearing deteriorated with age,especially at 12 months.Nlrp3 is one of the factors involved in sensorineural hearing loss and senile hearing loss.
