Observation of developmental changes of hair cell synaptic ribbons in C57 mice

Abstract:Objective To investigate the temporal characteristics of development of the synaptic ribbon in cochlear hair cells in C57 mice. Methods The number and distribution of presynaptic ribbons on the whole cochlear basilar mem-brane were examined and recorded by confocal microscopy with immunofluorescence in C57 mice at different develop-mental stages. ABRs were used to detect the time of hearing onset. Results Ctbp2 signals were present in the cytoplasm of inner and outer hair cells at day 0, continuing increasing at day 3 and reaching peak values (35.1 ± 3.4/inner hair cell and 21.0 ± 3.2/outer hair cell) at day 6. Subsequently, Ctbp2 signals tended to stabilize at 16-20/inner hair cell and 1-3/outer hair cell, with a gradual distribution shift to the basolateral wall of hair cells. In addition, ABR tests suggested that the average time of hearing onset in C57 mice was 13.2 days. Conclusions Synaptic ribbons in hair cells are present at birth, and continue to develop and mature after birth. The seemingly synchronized development of hearing and matura-tion of synaptic ribbons suggest that the latter may be a pre-condition for the former.
