STAT3 Expression during Cisplatin-induced Hair Cell Injury in the Lateral Line in Zebrafish Larvae

Abstract:Objective To develop a cisplatin-induced hair cell injury model using the lateral line system of zebrafish larvae and to detect the expression of STAT3 mRNA before and after cisplatin exposure. Methods Zebrafish larvae which were 5 days-post-fertilization were exposed to 250μM cisplatin for 3 hours. Yo-pro-1-and Terminal dUTP Nick-End-Label-ing (TUNEL-labeling) were used to examine hair cell densities and apoptosis, respectively. RT-qPCR and in situ hybridiza-tion were used to examine the expression level of STAT3. Results After being exposed to 250μM cisplatin for 3 hours, hair cell densities of the neuromast in the lateral line of zebrafish larvae were significantly decreased (P<0.001), and the num-ber of apoptotic cells significantly increased (P<0.001). Meanwhile, STAT3 expression was up-regulated (P<0.001). Con-clusion STAT3 may be involved in hair cell apoptosis during hair cell injury induced by cisplatin. While apoptosis is activat-ed at the same time, it indicates that STAT3 may be able to induce apoptosis.
