Melting curve quantitative technology in quantification of mitochondrial DNA 1555A>G heteroplasmic mutation

Abstract:Objective The melting curve quantitative technology was established using the TaqMan probe and com-pared with DHPLC and Sanger sequencing technologies in detecting mtDNA 1555A>G heteroplasmic mutation. Methods TaqMan probe melting curve technology was used to test aminoglycoside-induced hearing loss families and normal individu-als. The results were compared with our previous results using DHPLC and Sanger sequencing technologies. Results The level of mutant mtDNA 1555A>G was detected by TaqMan probe melting curve technology successfully in 50 samples, identifying one more case than Sanger sequencing, but three less than DHPLC. Conclusions The quantity of mutant mtD-NA 1555A>G copies ranging from 19% to 86% was accurately detected by the TaqMan probe melting curve technology used in this experiment.
