Abstract:Objective To study glutamate (glutamic acid, Glu) and gamma-aminobutyric acid (γ-GABA) expression levels in the cochlear nucleus of newborn guinea pigs with hyperbilirubinemia and effects of GABA and Glu on hearing dam?age. Methods Newborn guinea pigs were randomly divided into three groups:the control, and low and high bilirubin groups. Immunofluorescence was used to examine cochlear Glu and GABA levels;Western blotting analysis was used showing chang?es in cytochrome C expression. Results Compared with the control, GABA was significantly decreased (p<0.05) in both high and low bilirubin groups, whereas the levels of Glu were significantly increased (p<0.01). Western blotting showed significant?ly increased cytochrome C expression in bilirubin groups. Conclusion the dynamic changes of Glu and GABA in neonatal guinea pigs with hyperbilirubinemia reflects the status of neurons activities, suggesting that both may play important roles in bilirubin neurotoxicity affecting hearing. Cytochrome C increase in cytoplasm suggests that hyperbilirubinemia may induce early neurocyte aoptosis.
