Normal Morphology of Chick Columella During Development

Abstract:Objective Master normal morphology and establish a criterion of chick columella during its development, and find a method which can show the configuration of deep located columella in the whole mount. Method Collected E5-E19 chick embryos’heads, fixed with 4%PFA, stained cartilages with Alcian Blue, cleared tissue by methyl salicylate, and ob?served columella and other cartilages around by anatomy microscope. Results Columella came out on E7, keeping as 2 sepa?rate parts during E8-E10, displayed matured structure on E11(also other cartilages around it), on E12, columella’s configu?ration was clearer, While blood vessel’s shade came out on E13 and calcification began on E14. After E15, there were no mor?phology changes found, but transparence of connective tissue decreased and blood vessel’s shade increased. Conclusions Columella came out on E7 and showed matured morphology on E11(also other cartilages around it), and it is a good method for showing deep located columella in the whole mount to stain cartilages by Alcian Blue and clear tissue with methyl salicylate.
