Establishment and Optimization of an In Situ Whole Chick Embryo Hybridization Protocol

Abstract:Objective To report an experimental protocol for in situ hybridization in whole chick embryo with improved signal specificity and strength and reduced background noise for favorable conditions for future studies. Methods In situ hy?bridization of Bmp4(bone morphogenetic protein 4)in whole chick embryo was conducted and key factors affecting the testing process and optimal working conditions were investigated. Results Hybridization signals of high specificity and strength were obtained for Bmp4 with low background signals with optimized methodologies. Conclusions We have successfully established and optimized in situ hybridization of Bmp4 in chick whole embryo.
