Regulation of Epithelial Sodium Channels and Cochlear Function by Desmopressin

Abstract:Objective To investigate effects of desmopressin (DDAVP) on epithelial sodium channel (ENaC) expres-sion in the cochlea, and to study its possible role in the etiology of endolymphatic hydrops. Methods Twenty-four guinea pigs were divided into 3 groups. They received systemic injection of desmopressin for 0, 5 or 10 days respectively. The cochle-ae were examined by electrocochleography (ECochG), stained with HE and immunohistochemistry, and tested with Western blot. Results After being treated with DDAVP, ECochG showed mild endolymphatic hydrops in one guinea pig which was ver-ified by HE staining.α-ENaC was expressed in the stria vascularis, spiral ligament, Reissner’s membrane, organ of Corti, spiral limbus, spiral ganglions and cochlear nerve. Western blot showed that α-ENaC protein in the cochlea increased in DDAVP-treated guinea pigs (P<0.05). Conclusion Desmopressin increases the expression of α-ENaC in the cochlea, which may induce endolymphatic hydrops and affect function of the inner ear.
