Abstract:Objective Based on network pharmacology and molecular docking, this paper explores the potential molecular mechanism of Shangzhongxia Tongyong Gout formula in treating Rheumatoid Arthritis(RA),Systemic Lupus Erythematosus(SLE),Sicca syndrome(SS),Gout and other rheumatic immune diseases. Methods Using database to screen the active components and their targets of the Shangzhongxia Tongyong Gout formula, and predict the disease targets.Further screening the components-disease intersection targets, the key therapeutic targets and pathways. Finally, preliminary verification by molecular docking. Results The core components of the Shangzhongxia Tongyong Gout formula are baicalein,gentisin,wogonin,artemetin,β-sitosterol and stigmasterol, and the key targets are Tumor Necrosis Factor(TNF),Peroxisome proliferator-activated receptor gamma(PPARG),Interleukin(IL)-1β,Prostaglandin-endoperoxide synthase 2(PTGS2),Xanthine Dehydrogenase(XDH).GO enrichment and KEGG signal pathway analysis showed that the target protein of the Shangzhongxia Tongyong Gout formula involved in inflammatory reaction, circulation, metabolism and other biological processes was highly enriched, and it plays a therapeutic role on SLE,Gout,SS and RA through IL-17,TNF,toll-like receptor and C-type lectin receptor signal pathway. Conclusion There are common genes, proteins and biological processes among SLE,Gout,SS and RA diseases. The potential mechanism of the Shangzhongxia Tongyong Gout formula on such diseases can be realized by regulating immunity, metabolism and inflammation through multi-components such as Baicalein and Gentisin,multi-targets such as TNF and IL1B,and multi-pathways such as IL-17 pathway,TNF pathway and Toll-like receptor pathway.
