Research on the Sleep Improving Effect of Quansanqi Tablet

Abstract:Objective To observe the sleep-improving effects of Quansanqi tablet(PSQ).Methods SPF KM mice were divided into control group,positive control group,and multiple dose of PSQ groups.Sleep duration in mice was assessed using intraperitoneal injection of a threshold dose of pentobarbital sodium.The number of mice that fell asleep within 30 minutes was recorded after intraperitoneal administration of the maximum subthreshold hypnotic dose of pentobarbital sodium.Additionally,sleep latency within 60 minutes was measured following intraperitoneal injection of a hypnotic dose of barbital sodium.An insomnia rat model was established by intraperitoneal injection of PCPA,with normal,model,diazepam,and various PSQ dose groups being set up.Serum levels of γ-aminobutyric acid(GABA),dopamine(DA),and 5-hydroxytryptamine(5-HT)were determined,as well as mRNA expression levels of GABA and 5-HT in the hypothalamus.Results Compared with the control group,the sleep duration in the PSQ 0.60 and 1.20 g/kg groups was significantly prolonged(P<0.05),and the incidence of falling asleep were increased in the 1.20 g/kg group(P<0.05).All PSQ dose significantly shortened sleep latency(P<0.05).After PCPA modeling,compared with the normal group,the DA level in the serum of model group was significantly elevated(P<0.05),while GABA and 5-HT levels were significantly reduced(P<0.05),along with decreased mRNA levels of 5-HT and GABA in the hypothalamus(P<0.05).Compared with the model group,PSQ significantly lowered the DA level in the serum,increased 5-HT and GABA levels(P<0.05),and enhanced mRNA expression of 5-HT and GABA in the hypothalamus(P<0.05).Conclusion PSQ can significantly prolong sleep duration,increase sleep occurrence,and shorten sleep latency,possibly by regulating central neurotransmitter balance.
