Exploration of the Mechanism of Intervention of"Relaxation and Tranquility"Rehabilitation Concept in Spastic Hemiplegia Caused by Stroke Based on Inflammation and Oxidative Stress

Abstract:Objective To explore the key active ingredients and target pathways of Shujin Huanjing Yin in the treatment of spastic hemiplegia afterstroke,and investigate the mechanism of the"Relaxation and Tranquility"rehabilitation concept.Method The effective active ingredients,targets,and disease targets of Shujin Huanjing Yin were obtained through databases such as TCMSP,GeneCards,OMIM and Uniport.By conducting topology analysis and protein-protein interaction(PPI)analysis on drug disease intersection targets,key active ingredients and core targets were screened,enrichment analysis was used to obtain relevant pathways,and preliminary validation was conducted through molecular docking.Result There were a total of 99 active ingredients in Shujin Huanjing Yin,and 36 targets shared with stroke and spastic hemiplegia.Quercetin,mangiferin,kaempferol,naringenin,and β-sitosterol maybe were the key effective active ingredients.The core targets included interleukin-1 β(IL-1 β),protein kinase B1(Akt1),tumor necrosis factor(TNF),IL-6,tumor suppressor p53(TP53)and transcription factor AP-1(JUN).It mainly involved advanced glycation end product-receptor for advanced glycosylation end(AGE-RAGE)products pathway,IL-17 pathway,TNF pathway,lipid and atherosclerosis pathway,fluid shear force and atherosclerosis pathway.According to molecular docking,it can be seen that the docking result is good and feasible.Conclusion The treatment of post-stroke spastic hemiplegia with Shujin Huanjing Yin has the characteristics of multi-component,multi-target,and multi pathway,it may improve spastic hemiplegia after stroke by improving inflammation and oxidative stress after stroke,reducing pro-inflammatory cytokines and oxidative stress products,decreasing neuronal apoptosis,promoting tissue damage repair,protecting the blood-brain barrier,regulating lipid and energy metabolism.
