Exploration of the Repositioning Research and Development Model of Traditional Chinese Patent Medicines and Simple Preparations Indications Related to Multi-Dimensional Biological Network Under the Concept of Treating Different Diseases with Same Therapy

Abstract:The secondary development of Patent Chinese Medicines(PCMs)aims to achieve"two relative clarifications":relatively clarified pharmacodynamic material basis and relatively clarified pharmacological mechanisms.Clear clinical positioning is the foundation for realizing the clinical value of PCMs.Drug repositioning of PCMs refers to the process of expanding new indications or refining therapeutic advantages for marketed PCMs through further research.The fundamental guiding principle for PCMs repositioning is"treating different diseases with the same therapy",which at the microscopic level manifests as similarities in pharmacodynamic substances and mechanisms of action.This study proposes a three-stage repositioning R&D model:"TCM theoretical exploration-spectrum-effect-biological network analysis-clinical trial verification".First,guided by the"treating different diseases with the same therapy"concept,preliminary clinical positioning is established through TCM theoretical research.Then,formula similarity comparisons are conducted based on active ingredient studies,and multi-dimensional biological networks are constructed using multi-component,multi-target and multi-phenotype omics technologies to predict potential clinical advantages and screen efficacy indicators.Finally,confirmatory clinical trials are conducted.This model provides an effective strategy for establishing precise repositioning of PCMs with TCM characteristics.
