Study on the Mechanism of Notoginseng Total Saponins in Ameliorating Oxidative Stress Injury of Gastric Mucosa in Rats with Ulcer by Regulating the Nrf2/HO-1/NQO1 Pathway

Abstract:Objective To study the mechanism of notoginseng total saponins(NTS)in improving oxidative stress injury of gastric mucosa in rats with gastric ulcer based on the nuclear factor erythroid-2 related factor(Nrf)2/hemeoxy genase(HO)-1/NADPH quinineoxidoreductase(NQO)1 pathway.Methods SD rats were randomly divided into sham surgery group(SO group),model group(M group),NTS(18 mg/kg)group,ML385(Nrf2/HO-1 pathway inhibitor,20 mg/kg)group,NTS(18 mg/kg)+ML385(20 mg/kg)group,with 12 rats in each group.Except for the SO group,the other groups used acetic acid cauterization to establish gastric ulcer rat models.After group treatment with drugs,the area of gastric ulcer in rats were detected;the levels of oxidative stress factor in the gastric mucosal tissue,the serum levels of inflammatory related factors and gastric mucosal repair factor of rats were detected with kits;the expression of Nrf2/HO-1/NQO1 pathway related proteins in gastric mucosa tissue of rats was detected with Western blot method.Results Compared with the SO group,the gastric mucosal tissue of the M group had severe pathological damage,the levels of malondialdehyde(MDA),cyclooxygenase(COX)-2 and interleukin(IL)-6 were significantly increased(P<0.05),the levels of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD)and the expression of Nrf2,HO-1 and NQO1 proteins were significantly reduced(P<0.05).Compared with the M group,the pathological damage of gastric mucosal tissue was reduced in the NTS group,the area of gastric ulcers and the levels of MDA,COX-2 and IL-6 were reduced(P<0.05),and the levels of trefoil factor(TFF)1 and epidermal growth factor(EGF),GSH-Px and SOD,the expression of Nrf2,HO-1 and NQO1 proteins were increased(P<0.05);After adding ML385,the above pathological changes in the model rats were more severe,and ML385 could reduce the improvement effect of NTS on the above pathological changes in gastric ulcer rats.Conclusion NTS can prevent oxidative stress and inflammatory response in gastric ulcer rats by upregulating the expression of Nrf2/HO-1/NQO1 pathway proteins,thereby reducing gastric mucosal tissue damage and improving gastric ulcer symptoms.
