Abstract:Objective:In order to elucidate the mechanism of TMP inhibiting VSMC proliferation,we examined the AP-1 and PCNA protein expression in VSMC induced by PDGF.Methods:Cultured cells proliferating model of rats'VSMCs were established bv PDGF.Cultured cells were divided into three groups:control group、PDGF group and TMP(low dose, middle dose, high dose)group. The number of VSMC were accounted under microscope. The expression of activator protein-1(AP-1)and proliferating cell nuclear antigen(PCNA)were analysed by Western blotting respectively. Results:The proliferation of rat VSMCs induced by PDGF can be inhibited by TMP significantly in a dose-dependent manner.A remarkable decrease of AP-1 and PCNA expression were demonstrated in the TMP group compared with the model group. Conclusion:TMP inhibits VSMC proliferation associated with down regulation of AP-1and PCNA expression.
