Effect of NO in Hippocampus on Sleep-Wakefulness Cycle of Rats

Abstract:Objective:To investigate the effects of nitric oxide (NO) in hippocampus on sleepwakefulness cycle of rat.Methods:Stereotaxic,microinjection,polysomnography (PSG) and nucleus spile.Results:N-nitro-L-arginine (L-NNA,2.0 μg),an inhibitor of nitric oxide synthase (NOS),increased slow wave sleep (SWS) and decreased wakefulness.Sodium nitrioprusside(SNP,0.2 μg),the donor of NO,not only decreased wakefulness but also abolished the sleep-promoting effect of L-NNA.L-arginine (L-Arg,0.4 μg),a precursor of NO,had no direct effect on sleep and wakefulness,but blocked the effect of L-NNA.Conclusion:Hippocampus plays an important role in regulating sleep and wakefulness,and NO in hippocampus can promotes wakefulness.
