Study of Modulating Effect of ZuoGui Pill on Rat Thymocytes Proliferation

Abstract:Objective:To investigate the accommodation and the molecular mechanism of Zuogui Pill (ZGP) on the proliferation and immuno function of rat thymocytes (MSTM) in vitro. Method:Thymocytes were divided into three groups : (A) control group ; ( B ) model group ; ( C ) ZGP group. The effect ZGP with corticosterone on the proliferation of thymocytes was detected by MTT. Cell cycle was measured by PI staining and FCM. The expression of p16, cdk4, IL-2, IL-2R were measured by Real-time PCR and Western Blotting. Resuhs:Corticosterone had some inhibitory effect on thymocytes, arrested cell cycle at G_1 phase, up-regulated the expression of p16, down-regulated the expression of IL-2, IL-2R.Comparing with model group, ZGP group showed higher expression of IL-2 and IL-2R and lower expression of p16 (P < 0. 05 ). Conclusions : The possible mechanism of immuno-regulation effect of ZGP is to regulate the expression of p16, IL-2, IL-2Rα in MSTM, postpone the thymus dependented immune functional deterioration induced by corticosterone.
