Research status of targeted therapy for gallbladder cancer

Abstract:Gallbladder cancer(GBC)is the most common malignant tumor of the biliary system,with a highly aggressive and poor prognosis profile.Due to its insidious onset,most patients are already in the middle or late stages at the time of diagnosis,and have few opportunities for surgical resection.This article summarizes the current research of targeted therapy for GBC,foucsing on the key signaling pathways-related targets,such as the ErbB receptor,VEGF/VEGFR pathway,MAPK pathway,and PI3K/Akt/mTOR pathway.Some targeted drugs have shown certain efficacy in clinical trials,but there are still problems such as tumor heterogeneity,molecular mechanism of drug resistance,and detection techniques bottleneck.Bioinformatics methods have identified emerging potential targets and their drug candidates such as PTBP3 and WEE1,which are still in the basic research stage.Therefore,GBC still faces many challenges,and future research needs to focus on the integration of multi-omics technology,the optimization of combination treatment regimens,the innovation of high-sensitivity detection methods,and international multi-center collaboration to promote therapeutic breakthroughs.
