Abstract:Colorectal cancer(CRC)is the third most common malignant tumor worldwide and the second leading cause of cancer-related deaths globally.Despite recent improvements in the comprehensive treatment of malignant tumors, the prognosis for metastatic colorectal cancer(mCRC)remains poor.Notably,5%of mCRC cases exhibit alterations in the Erb-B2 receptor tyrosine kinase 2(ERBB2).ERBB2,commonly known as human epidermal growth factor receptor 2,is a member of the human epidermal growth factor receptor family and belongs to the protein tyrosine kinase class. In addition to being recognized as a therapeutic target in the treatment of gastric and breast cancer,it is also considered crucial in the management of colorectal cancer. In this review,we describe the molecular biology of ERBB2 from the perspective of biomarkers for targeted therapy in mCRC,new targeted treatment methods, and ongoing clinical trials. This may help to establish new treatment standards for patients with ERBB2-positive mCRC.
