Research of extracellular matrix in EGFR-TKI resistance in non-small cell lung cancer

Abstract:Epidermal growth factor receptor (EGFR)-Tyrosine kinase inhibitors (TKIs) significantly improve the prognosis of patients with EGFR mutant non-small cell lung cancer ( NSCLC) .Despite the efficacy of EGFR-TKI is significant,the drug resistance is inevitable and the mechanisms of drug resistance in most patients are not clear.The extracellular matrix ( ECM ),as an important component of the tumor microenvironment,plays a crucial role in regulating drug resistance,invasion,and metastasis of tumors.This review will focus on the composition and function of ECM,explore the effect and related mechanism of ECM on EGFR-TKI drug resistance,and review the relative studies on overcoming EGFR-TKI resistance through targeting ECM in NSCLC,providing new strategies for overcoming drug resistance.
