Application of organoids in immunotherapy for triple-negative breast cancer

Abstract:Organoids are a new type of in vitro 3D cell culture model.Tumor organoids can maintain the same genotype and biological characteristics as the original tissue,and can stably maintain a high degree of heterogeneity of tumor cells.Breast cancer is the most common malignancy worldwide and has complex genetic and pathological features.Tumor resistance,recurrence and metastasis are the main reasons for treatment failure in the vast majority of breast cancer patients.Although immunotherapy has changed the way cancer is treated,there is a lack of efficient and robust detection platforms.Immunotherapy requires the detection of not only cancer cells,but also the tumor microenvironment( TME) .Organoids can capture the full heterogeneity of breast cancer by reproducing a patient's original tumor in terms of histopathology,hormone receptor expression,HER-2 status,copy number variation,and sequence changes.Organoid technology can be very helpful in revealing the causes of tumor heterogeneity,evolution,and metastasis,as well as evaluating the efficacy of drugs.This article will discuss the basic principles of organoids and their application and progress in breast cancer immunotherapy.
