Predictive value of RGS16 and HMGB1 protein expression in recurrence after cold knife conization for HSIL of the cervix

Abstract:Objective To investigate the predictive value of G protein signal regulator(RGS)16 and high mobility group protein B1(HMGB1)expression in the recurrence of high-grade cervical squamous intraepithelial lesions(HSIL)after cold knife conization(CKC).Methods A total of 238 patients with cervical high grade squamous intraepithelial lesion(HSIL)who underwent CKC and whose postoperative pathological margins were negative in Lianyungang Hospital Affiliated to Xuzhou Medical University from January 2021 to December 2022 were selected.Lesion tissues were collected during the operation.The protein expressions of Regulator of G protein Signaling 16(RGS16)and High mobility group protein B1(HMGB1)were detected by immunohistochemistry.After 1 year of follow-up,the patients were divided into a recurrence group and a non-recurrence group based on whether they had recurrence within 1 year after surgery.The expression of RGS16 and HMGB1 proteins between the two groups was compared,and univariate analysis was performed on the clinical data of the two groups.Factors with statistically significant differences in univariate analysis were included in the multivariate Logistic regression analysis to screen for factors influencing postoperative recurrence in HSIL patients.Receiver operating characteristic(ROC)curves were plotted,and the area under the curve(AUC)was used to evaluate the predictive value.Results There was no loss to follow-up during the 1-year follow-up period.Among the HSIL patients,24 cases had recurrence,with a recurrence rate of 10.08%,while the remaining 214 cases had no recurrence.Univariate analysis showed that the number of pregnancies,involvement of cervical glands,persistent human papillomavirus(HPV)infection after surgery,proportion of positive liquid-based cytology after surgery,positive expression rates of RGS16 and HMGB1 proteins were higher in the recurrence group than in the non-recurrence group(P<0.05).Multivariate Logistic regression analysis showed that positive expression of RGS16 protein and HMGB1 protein were factors influencing postoperative recurrence in HSIL patients(P<0.05).ROC curve analysis showed that the AUC values for predicting postoperative recurrence in HSIL patients were 0.740 for RGS16 protein,0.790 for HMGB1 protein,and 0.818 for the combination of both proteins(P<0.05),with a higher AUC value for the combination(P<0.05).Conclusions Positive expression of RGS16 protein and HMGB1 protein can be used to predict the risk of postoperative recurrence in HSIL patients,and the combination of both proteins has a higher predictive value.
