Expression of LCAT in endometrial carcinoma and its clinical significance:A study by bioinformatics analysis

Abstract:Objective To investigate the association of the expression of lecithin cholesterol acyltransferase(LCAT)gene with clinicopathological parameters and prognosis in endometrial carcinoma(EC)patients.Methods The expressions of LCAT in EC were analyzed by UALCAN,GEO,and TCGA database.The correlations between LCAT expression and clinicopathologic characteristics were analyzed by the t-test(or Wilcoxon test)and Chi-square test.Kaplan-Meier method and Cox regression analysis were used to evaluate the relationship between clinicopathologic features and overall survival(OS)of EC patients.The expression of LCAT mRNA in 15 endometrial carcinoma tissues and 15 normal endometrial tissues were also assessed by Real-time quantitative polymerase chain reaction(RT-qPCR).Results According to UALCAN,GEO,and TCGA database analysis,the expression of LCAT was significantly decreased in endometrial carcinoma compared to normal endometrium(P<0.01).The results of clinical sample testing were consistent with the results of database analysis.Lower LCAT expression was significantly related to age,tumor grade,histological type,and myometrial invasion(P<0.05).Kaplan-Meier survival analysis revealed a significantly shorter overall survival in the low expression group of LCAT compared to the high expression group(P<0.01).Furthermore,Cox regression demonstrated that LCAT expression could serve as an independent predictor for poor outcome of EC(P<0.05).Conclusions LCAT expression may be associated with the development of EC,and its low expression might be used as a potential prognostic molecular marker of poor survival in EC.
