Research progress of fusion genes in salivary gland tumors

Abstract:Salivary gland tumors are common oral and maxillofacial tumors with heterogeneity.The treatment options for inoperable,recurrent,and metastatic patients of salivary gland tumors are very limited,and the discovery of fusion genes opens up a new way to their diagnosis and treatment.The main targets of fusion genes encoding novel fusion oncoproteins or ectopic expressed normal/truncated oncoproteins are transcriptional coactivators,tyrosine kinase receptors,and transcription factors involved in growth factor signaling and cell cycle regulation,some of which or pathways activated by these targets are targetable.With the further step into the era of precision medicine and targeted therapy,more comprehensive molecular analysis will provide new insights into the diagnosis and molecular pathogenesis of various salivary gland tumors,and provide more treatment options for patients with poor response to conventional treatment.This article reviews the types,functional roles,and targeted drugs of fusion genes in benign and malignant salivary gland tumors,aiming to provide a new direction for the diagnosis,mechanism,and clinical treatment of fusion genes in salivary gland tumors.
