The Emodin inducted human breast cancer MCF-7cell apoptosis and activated JNK signal transduction pathways

Abstract:Objective To explore the Emodin impact on human breast cancer MCF-7 cell proliferation and apoptosis, as well as related to JNK signal pathway. Method Application of inverted phase contrast microscope observed human breast cancer MCF-7 cell growth in vitro, proliferation of MCF-7 cell was detected by MTT meth-od, and apoptosis of MCF-7 cell was detected by flow cytometer. Using Western blot and immunohistochemistry methods detected expressions of JNK and activator protein 1(AP-1) related apoptosis. Results The Emodin could reduce human breast cancer MCF-7 cell proliferation vary with time and the concentration respectively, and there had a statistically significance (P <0. 05). The Emodin reduced human breast cancer MCF-7 cell early ap-optosis phenomenon, in a concentration dependence way, with a statistically significant (P <0. 05). Different con-centration of Emodin could enhance expression of JNK signal downstream of AP-1 gene related apoptosis in human breast cancer MCF-7 cell, and there had a statistically significance (P < 0. 05). Conclusions The Emodin could reduce proliferation capacity of human breast cancer MCF-7 cell in vitro, promote early apoptosis, enhance expressions of JNK and AP-1 related apoptosis in human breast cancer MCF-7 cell. The Emodin could inhibit hu-man breast cancer MCF-7 cell proliferation, promote apoptosis by activation of JNK signal transduction pathways.
