Analysis on the correlation between MTHFR and PROC gene polymorphisms and the occurrence of venous thromboembolism in patients with malignant tumors

Abstract：Objective To analyze the correlation between methylenetetrahydrofolate reductase(MTHFR)and protein C(PROC)gene polymorphisms and the occurrence of venous thromboembolism(VTE)in patients with malignant tumors.Methods The clinical data of 227 patients with malignant tumors who were admitted to Affiliated Cancer Hospital of Xinjiang Medical University from November 14,2023 to February 14,2024 were retrospectively analyzed.According to the occurrence of VTE,the patients were divided into VTE group(47 cases)and non-VTE group(180 cases).The polymorphisms of PROC gene(rs 199469469 locus)and MTHFR gene(rs1801133 locus)and the other clinical data were compared between the two groups.Multivariate logistic regression was used to analyze the factors affecting the occurrence of VTE in patients with malignant tumors.Results The level of D-dimer and the proportion of the patients with a history of diabetes in the VTE group were higher than those in the non-VTE group,and the differences were statistically significant(P＜0.05).There was a significant difference in the genotype distribution of rs 1801133 locus of MTHFR gene between the two groups(P＜0.05).The frequency of A allele of rs1 801133 locus of MTHFR gene in the VTE group was significantly higher than that in the non-VTE group[53.19％(50/94)vs 38.89％(140/360),x2=3.945,P=0.047].There was no significant difference in the genotype of rs199469469 locus of PROC gene between the two groups(P＞0.05).The results of multivariate logistic regression analysis showed that higher D-dimer level[OR(95％CI)=1.436(1.213-1.696)],a history of diabetes[OR(95％CI)=2318(1.125-6.808)],AA genotype of rs1801133 locus of MTHFR gene[OR(95％CI)=1.927(1.459-3.751)]were independent risk factors for promoting the occurrence of VTE in patients with malignant tumors(P＜0.05).Conclusion The polymorphisms of rs1801133 locus of MTHFR gene are correlated with the occurrence of VTE in malignant tumor patients,but the polymorphisms of rs199469469 locus of PROC gene are not significantly correlated with the occurrence of VTE.
