Analysis on the effect of TGF-β signaling pathway on osteogenic differentiation of odontogenic myxoma mesen-chymal stem cells

Abstract:Objective To explore the effect of transforming growth factor-β(TGF-β)signaling pathway on osteogenic differentiation of odontogenic myxoma(OM)mesenchymal stem cells(OMMSC).Methods Two patients with OM who were admitted to Stomatological Hospital Affiliated to Medical School of Nanjing University from 2018 to 2020 were selected.The patients'partial lesion specimens were obtained through surgery for OMMSC isolation and culture.In addition,5 young patients who received dental implant therapy in the hospital were selected to extract bone marrow from the jaw bone for jaw bone mesenchymal stem cells(JBMSC)isolation and culture.The cells were identified by cell proliferation assay,flow cytometry and alizarin red S staining.After 7 days of osteogenic culture,real-time fluores-cence quantitative polymerase chain reaction(RT-PCR)was used to detect the TGF-β signaling pathway and the expression of osteogenesis-related genes.The effects of small molecule drug SB431542 and TGF-β1 interventions on osteogenic differentiation ability of OMMSC and JBMSC were observed.Results OMMSC and JBMSC obtained by isolation had spindle-shaped morphology,and expressed the surface markers of mesenchymal stem cells,and had osteogenic differ-entiation ability,which were consistent with the characteristics of mesenchymal stem cells.Compared with JBMSC,OMMSC had stronger proliferation ability.The results of RT-PCR showed that TGF-β/Smad signal-related factors were highly expressed in OMMSC(P<0.05),and the expression of osteogenesis-related factor osteopontin(OPN)was decreased(P<0.05).The results of alizarin red S staining showed that SB431542 could significantly promote the osteogenic differ-entiation of OMMSC,while TGF-β1 inhibited the osteogenic differentiation of OMMSC.Conclusion In this study,OMMSC are successfully isolated,and TGF-β signal-related factors are highly expressed in OMMSC,and inhibiting TGF-β signal transduction pathway can enhance the osteogenic ability of OMMSC,which provides reference for improving bone defects in patients with OM.
