Effect of lentivirus vector-mediated TSPY1 gene on proliferarion of SMMC7721 and Huh7 hepatocellular car-cinoma cells
Abstract：Objective To investigate the effects of lentivirus vector-Testis-specific Protein Y-encoded 1 (TSPY1)on the proliferation ability of SMMC7721 and Huh7 hepatocellular carcinoma cells .Methods The lentivir-us vector was constructed and transfected into the SMMC 7721 and Huh7 cells.The expression of TSPY1 mRNA and protein was detected by quantitative RT-PCR and western blot .Cell Counting Kit-8 ( CCK8 ) assay was performed to assess the transfected cells proliferation .Results The expression of TSPY1 both in mRNA and protein levels were significantly increased in the SMMC 7721 and Huh7 cells transfected with lentivirus encoding TSPY 1 cDNA ( P <0.01 ) .CCK8 assay indicated that TSPY 1 potentiates cell proliferation both in SMMC 7721 and Huh7 cells over-ex-pressed TSPY1, comparing to the control cells .Conclusion The transfection of lentiviralvector-mediated TSPY1 gene can obviously up-regulate the expression of TSPY 1 mRNA and protein and promote the proliferation of SMMC7721 and Huh hepatocellular carcinoma cells .
