Exploring the mechanism of Ginkgo leaf extract in the treatment of benign paroxysmal positional vertigo based on network pharmacology

Abstract:Objective To explore the mechanism of Ginkgo leaf extract in the treatment of benign paroxysmal positional vertigo(BPPV)through network pharmacology research methods.Methods First,the effective active components and target genes of Ginkgo leaf were obtained from the TCMSP database.The target genes of BPPV were retrieved from the GeneCards database.Then a"drug-effective component-target"network was constructed to analyze the key active components.Using the STRING database,a protein-protein interaction(PPI)network was built to investigate the interaction relationships among related proteins.Finally,the core targets were subjected to GO functional enrichment and KEGG pathway enrichment analysis via the Metascape database.Results The effective active ingredients of Ginkgo leaf were screened,yielding 27 compounds and 253 related targets which were converted to gene symbols and reduced to 112 after removing duplicates.There were 1,600 target genes associated with BPPV,with 47 overlapping genes related to Ginkgo leaf drug.GO functional analysis primarily involved responses to inorganic substances,hypoxia,hormones,positive regulation of cell migration,and reactions to lipopolysaccharides.KEGG pathway analysis mainly covered lipid and atherosclerosis,cancer pathways,the AGE-RAGE signaling pathway in diabetic complications,activation of chemical carcinogens,and proteoglycans in cancer.Conclusion The potential mechanism of Ginkgo leaf extract in treating BPPV is as follows:quercetin,kaempferol,luteolin,and isorhamnetin may be the primary effective bioactive components responsible for its efficacy.IL-6,MYC,VEGFA,HIF1 A,NFKBIA,and FOS could be potential targets of Ginkgo leaf in treating BPPV,exerting biological effects such as anti-inflammation,antivirus,antitumor,antioxidation,antiplatelet aggregation,and immune response activities.
