Efficacy of sequential anthracycline-based chemotherapy followed by dual-target therapy combined with docetaxel in the treatment of HER2-positive breast cancer

Abstract:Objective To evaluate the efficacy of anthracycline-based sequential dual-target therapy combined with docetaxel in the treatment of human epidermal growth factor receptor 2(HER2)-positive breast cancer.Methods This study is a prospective controlled study.A total of 128 HER2-positive breast cancer patients admitted to Xingtai Central Hospital from March 2023 to March 2025 were selected and randomly divided into the observation group(n=64,receiving sequential anthra-cycline-based chemotherapy followed by dual-targeted therapy combined with docetaxel in 21-day cycles for a total of 8 cycles)and the control group(n=64,receiving dual-targeted therapy combined with docetaxel in 21-day cycles for a total of 4 cycles)using a random number table method.Clinical efficacy,tumor marker levels,cardiac function parameters,and the incidence of adverse reactions were analyzed and compared between the two groups.Results The objective response rate in the obser-vation group was 70.31%(45/64),higher than that in the control group(46.88%(30/64))(x2=7.245,P<0.05).After treatment,the levels of carcinoembryonic antigen and carbohydrate antigen 153 decreased in both groups,with lower values observed in the observation group((5.08±1.02)ng/mL,(28.17±7.45)U/mL,respectively)compared to the control group((9.95±1.63)ng/mL,(46.42±10.52)U/mL,respectively)(t=20.262,11.326,both P<0.05).After treatment,there were no statistically significant differences in left ventricular ejection fraction and N-terminal pro-B-type natriuretic peptide between the two groups(t=1.048,0.530,both P>0.05).The incidence of alopecia in the observation group was 14.06%(9/64),lower than that in the control group(34.38%(22/64)),while the incidence of lower extremity edema in the observation group was 40.62%(26/64),higher than that in the control group(14.06%(9/64)(x2=7.194,11.365,both P<0.05).Conclusion Sequential anthracycline-based chemother-apy followed by dual-target therapy combined with docetaxel is effective in treating HER2-positive breast cancer.It effectively reduces the levels of tumor markers,decreases the incidence of alopecia,and does not increase cardiotoxicity,although it notably increases the incidence of lower extremity edema.
